Introduction |
Lysozyme is a glycoside hydrolase, which is composed of 18 kinds of 129 amino acid residues, with 4 S-S bonds and molecular weight of 14307. Lysozyme is a kind of hydrolase, which can dissolve the cell wall of Gram-positive bacteria, has strong killing effect and can also be used for crystallization Protein lysozyme is widely used in food preservation (especially cheese and sake production), medicine (tablets, capsules, eye drops, throat drops), daily chemicals (toothpaste, cosmetics), baby food (breast milk milk powder) and scientific research. |
Function |
Lysozyme can effectively hydrolyze peptidoglycan in bacterial cell wall, and the hydrolysis site is the β - 1.4 glycosidic bond between the 1-carbon atom of n-acetyl-cell wall acid (NAM) and the 4-carbon atom of N-acetylglucosamine (NAG). Peptidoglycan is the main component of bacterial cell wall. It is composed of Nam, NAG and peptide "tail" (usually four amino acids). Nam and nag are connected by β - 1.4 glycosidic bond, while peptide tail is connected to the third carbon atom of Nam through d-lactacyl carboxyl group. Peptide tails are connected by peptide "bridge" (peptide bond or a few amino acids). Nam, nag, peptide "tail" and peptide "bridge" are composed of them Peptidoglycan is a kind of multi-layer network structure, which is the skeleton of cell wall. Any chemical bond break in the above structure can lead to the damage of bacterial cell wall. For Gram-positive bacteria (G +), such as Micrococcus luteus, Bacillus subtilis or Micrococcus lysozyme, the content of peptidoglycan in cell wall of Gram-positive bacteria (G +) is different from that of Gram-negative bacteria (G -), such as Escherichia coli, proteus, dysentery bacilli and pneumoniae. Almost all the cell walls of G + bacteria are composed of peptidoglycan, while only the inner wall layer of G-bacteria is peptidoglycan+ The cell wall of bacteria is stronger than that of G-bacteria. |
Application |
1. Preservation
Lysozyme can decompose Bacillus subtilis and radiation-resistant Micrococcus in Gram-positive bacteria by decomposing the cell wall of bacteria, and also can dissolve Gram-negative bacteria such as Escherichia coli, Proteus vulgaris and Vibrio parahaemolyticus to a certain extent. It is widely used in the antisepsis of aquatic products, meat food, cake, sake, cooking wine and beverage.
2. Dairy processing
Adding a certain amount of lysozyme in fresh milk or milk powder not only has the function of antisepsis and preservation, but also can kill the intestinal spoilage cocci, increase the intestinal anti infection ability, promote the proliferation of intestinal Bifidobacterium, so as to promote the digestion and utilization of casein, which is commonly used in the addition of infant dairy products and food products.
3. Pharmaceutical industry
Lysozyme, as a non-specific immune factor existing in human body fluids and tissues, can be used as a natural anti infective substance with bactericidal effect. It has antibacterial, antiviral, hemostatic, detumescence and analgesic effects, and can accelerate the recovery of tissue function. It is used for chronic rhinitis, acute and chronic pharyngitis, oral ulcer, varicella, herpes zoster and verruca plana. It can also be combined with antibiotics to treat various bacterial and viral infections.
4. Daily chemical industry
The bacteriostasis principle is safe and harmless to human body. Adding to the mask and skin care products can play a role in keeping bacteria and keeping fresh, and also has anti-inflammatory effect on the skin. Added to toothpaste, it can promote swelling and recovery of gums.
5. Bioengineering
Lysozyme has the function of destroying the structure of bacterial cell wall, so lysozyme is an essential tool enzyme for cell fusion in genetic engineering and cell engineering.
